Brown spots, a milky precipitate (loss of vacuum) or no longer glowing tubes are immediately visible. More difficult to determine are "slowly dying tubes", in which the listening experience slowly changes. With aging preamp tubes, microphones, hissing and scratching noises creep in.
NOS means New Old Stock. These are new goods as old stocks.
The easiest option would be to look it up in the user manual or online with the type number. If these cannot be found, only a look at the defective tube will help. The type, manufacturer and tube socket can often be read off here.
